Circular RNA circLMO7 acts as a microRNA-30a-3p sponge to promote gastric cancer progression via the WNT2/β-catenin pathway

Abstract Background Gastric cancer (GC) is one of the most common malignant tumors worldwide. Currently, overall survival rate GC still unsatisfactory despite progress in diagnosis and treatment. Therefore, studying molecular mechanisms involved vital for CircRNAs, a type noncoding RNA, have been proven to act as miRNA sponges that can widely regulate various cancers. By this mechanism, circRNA at genetic level by releasing from inhibiting its target genes. The WNT2/β-Catenin regulatory pathway canonical signaling pathways tumors. It not only promote development but also provide energy tumor growth through cell metabolism (such glutamine metabolism). Methods Through RNA sequencing, we found hsa_circ_0008259 (circLMO7) was highly expressed tissues. After verifying circular characteristics circLMO7, determined downstream (miR-30a-3p) circLMO7 pull-down luciferase reporter assays. We verified effect miR-30a-3p on cells series functional experiments, including colony formation, 5-ethynyl-2′-deoxyuridine Transwell Western blot immunofluorescence analyses, WNT2 gene further confirmed circLMO7-miR-30a-3p-WNT2 axis could GC. In addition, measurement related metabolites metabolism. metastasis vivo establishment nude mouse models. Finally, demonstrated HNRNPL bind flanking introns exons cyclization. Results CircLMO7 acted sponge affecting proliferation, migration invasion cells. Moreover, animal results showed vivo. affect biological function. proved self-cyclization circLMO7. Conclusions promotes inhibitory WNT2.
